Modified Acyl Carrier Proteins

The Wisconsin Alumni Research Foundation (WARF) is seeking commercial partners interested in developing acyl carrier proteins modified with a non-radioactive label.
OVERVIEW
Acyl carrier protein (ACP) is a small protein that functions as an acyl carrier. It is involved in fatty acid biosynthesis and many other important biological processes in both eukaryotes and prokaryotes. ACP is difficult to obtain in a functional state from recombinant systems, primarily because posttranslational modification is required for proper protein functioning.
THE INVENTION
UW-Madison researchers have developed purified apo-, holo- and acylated-acyl carrier proteins modified with a non-radioactive label that is covalently bonded to a modified tyrosine residue within the protein. To generate the labeled proteins, an amino group is first introduced into a tyrosine residue of apo-ACP. Next, a fluorescent molecule is covalently bonded to the amino group. The labeled apo-ACP is then converted to holo-ACP and subsequently to acylated-ACP.

  • Investigating reactions involving or mediated by ACPs
KEY BENEFITS
  • Label is catalytically silent -- it does not interfere with binding of enzymes to ACP molecules.
  • Addition of label to tyrosine residue is very specific.
  • Enables interactions of ACPs to be examined in greater detail, with greater ease, and with greater precision than current radiolabeled ACPs
  • Involves no time-consuming and waste-generating radioactivity
