Faster, Cheaper DNA Synthesis

The Wisconsin Alumni Research Foundation (WARF) is seeking commercial partners interested in developing a method for creating hundreds of thousands of oligonucleotides at dramatically lower cost.
OVERVIEWSynthesizing DNA and genes is an important tool in biotechnology. It has become commonplace to synthesize short DNA chains, called oligonucleotides, on a substrate and use these as building blocks to assemble longer DNA sequences.

While the process can be automated, synthesizing long genes remains expensive. Also, current methods produce extremely small amounts of DNA, which makes physical handling and dilution a challenge.
THE INVENTIONUW–Madison researchers have developed a high yield method for synthesizing oligonucleotides with defined sequences and soluble in solution.

In the new method, DNA starting material is synthesized in parallel on a microarray, amplified via polymerase chain reaction (PCR) and cleaved. The cleaved fragments then can be used to assemble a double-stranded DNA target sequence.

Hundreds of thousands of oligonucleotides can be synthesized and assembled into many individual genes by this process.
