The Invention
UW-Madison researchers have generated a new human GATA2 deficiency model. The model was generated using CRISPR-Cas9 targeting to deactivate one copy of the GATA2 gene in HUDEP-2 cells. This cell line, coined HUDEP GATA2 low cells or HUG2low for short, exhibits an ~60% reduction in GATA2 protein. This reduction leads to a substantial disruption in the transcriptome (amalgamated pattern of gene expression) of these HUG2low cells. Restoring GATA2 to near normal levels in HUG2low cells reverses the aberrant pattern of gene expression.
