The Invention
UW-Madison researchers have developed an easy-to-use, robust, and generalizable method for ultrahigh-throughput single-cell genetic analysis of microbial populations using droplet microfluidics. Their method, coined Droplet Targeted Amplicon sequencing (DoTA-seq) uses a simple microfluidic droplet-marker to encapsulate single microbial cells into millions of picoliter-sized hydrogels, allowing multi-step cell lysis procedures. DoTA-seq leverages a multiplexed PCR reaction to attach unique barcodes to the targeted amplified loci of each cell, providing flexibility to the number and type of loci targeted for sequencing.
