RETRACTABLE ION GUIDE, GRID HOLDER, AND TECHNOLOGY FOR REMOVAL OF CRYOGENIC SAMPLE FROM VACUUM

UW-Madison researchers have developed an improved sample handling system for the preparation of samples to be analyzed using cryo-electron microscopy (cryo-EM). The system includes a retractable grid holder that compatible with existing EM sample grids. In operation, the sample is deposited on the EM grid and encased in an amorphous solid. Once deposited and coated, the grid holder can be retracted for analysis while maintaining vacuum. This sample handling system reduces exposure to contamination and other environmental factors (e.g., elevated temperatures), which can destroy the sample or lead to inaccurate analysis. Ultimately, this invention reduces sample preparation time and costs associated with handling.
