The Invention
UW-Madison researchers disclose a new method for differentiating brain mural cells – encompassing pericytes and vascular smooth muscle cells – from hPSCs. The inventors used their previously disclosed protocol (P170340US02) to derive a neural crest intermediate, and subsequently overexpress the Notch3 intracellular domain in neural crest. The result is acquisition of a brain mural cell-like fate, which the inventors confirmed via increased expression of key mural cell genes.
